Yes!! I do it at least twice a year. Nothing worse than having a flat and not being able to access your spare in the middle of nowhere.

I always keep it that way to avoid problems with the valve stem when getting high centered in a mud hole. Also, it makes me drop the spare and actually take a good look at it so that I can verify it's still in good shape.
